Gun Star Heroes (Genesis) would have to take my "best of all time" platformer pick - as long as you play it in 2 player mode, sadly it's never been re-released on PC.

I know this is the retro gaming forum, but there's heaps of modern platform games around with Win 7 compatible releases (that you don't need to install emulators for) that get overlooked a lot, so I present to you:

Drac's (Win 7 compatible) Retro Inspired Platforming Playlist:

Bionic Commando Rearmed

A fantastic remake of a true classic.
You can't go wrong with this. Get it on Steam.

This is the best 3D platformer of the PS2/Xbox/GC era, best of all
there is a native PC version. Snag it on Steam.

Braid is slow paced with fantastic time mechanics and great puzzles.
Get it on Steam.

Rocket Knight

Yay more Rocket Knight! Sadly not available via Steam in Australia (yet). Link to
USA Steam Store Page - You won't be able to purchase this from Australia right
now, I hope it's released here soon though.

Earthworm Jim 1 & 2

You can't go wrong when you give a space worm a raygun. Get it on GOG.

Duke Nukem Manhattan Project

A post Duke Nukem 3D game that was actually released!... Get it on GOG.

Charming physics based platform game.
You play a blob, no, seriously, it's good. Grab it on Steam.

Oddworld: Abe's Oddysee

Had trouble finding a trailer for this one.
Another great classic off-beat platformer made avaliable on GOG.

Another World

This is the 15th anniversary edition with higher resolution graphics.
Get it on GOG

Shinobi III

Classic ninja action! Avaliable at Gamers Gate.

N: The Way of The Ninja

Free for PC, fun, fluid and can get really hardcore.
Download or Play The Way of The Ninja Here

(Yes, that's the name of the game, V six times)

Very quirky but great retro themed platformer.
Download the demo or purchase from their website.

Very well polished physics based action & puzzle platformer, has online co-op!
Grab it on Steam.

Oh and one upcoming game to watch for: FEIST
. . . .

Super Metroid (SNES) Samus = the original kick-ass heroine.
Castlevania SotN (PS) "Psh. What is a man but a miserable pile of secrets?"
MegaMan X (SNES) I got to play Zero! To this day the only MM I ever won! (Except the Battle Network games, which are action/RPG, and therefore don't count.)

One of my favorite's was Gex (PS). Merely a typical platformer, but some of the one-liners were priceless.

Another that I wasted countless hours on was Space Taxi on the C64. Not because it was outstanding, but because it was one of the few games that I had at that time..... oh the simple days.
